The Poly(I:C) sodium component is a synthetic double-stranded RNA analog that agonizes TLR3 and the retinoic acid-inducible gene I receptor (RIG-I and MDA5); it can act as a vaccine adjuvant that boosts innate and adaptive immune responses and triggers apoptosis in cancer cells[1][2]. The Kanamycin component is an orally active antibacterial agent effective against both Gram-negative and Gram-positive bacteria, working by binding the 70S ribosomal subunit to block translocation and cause miscoding; it is notably active against Mycobacterium tuberculosis, including drug-resistant strains, and against Klebsiella pneumoniae, and can be used in research on tuberculosis and pneumonia[3]. In Vitro
In WM793, WM278, WM239A, WM9 and 1205Lu cells, Poly (I:C) sodium (20 ng/mL; 24 hours) strongly lowers viability from 100% in controls to 20%–50% within 24 hours[1]. In 1205Lu cells, Poly (I:C) sodium (200 ng/mL; 24 hours) induces apoptosis[1]. Poly (I:C) sodium (3 ng/mL; 24 hours; 1205Lu cells) triggers IFN-β expression in melanoma cells. Silencing RIG-I and MDA-5 showed that Poly (I:C) sodium-induced IFN-β expression required RIG-I and MDA-5, respectively, and that IPS-1 was also required[1]. Active subunits of caspase-9 and caspase-8 are revealed in melanoma cells by Poly (I:C) sodium (5 ng/mL; 24 hours; 1205Lu cells)[1]. Kanamycin (0.1-100 μg/mL; 2 weeks) shows good antibacterial activity against various mycobacterial strains in vitro (MIC=1-5 μg/mL)[3].
In Vivo
In NOD/SCID immunodeficient mice injected with 1205Lu cells, Poly (I:C) sodium treatment inhibits tumor growth, and human DNA levels are 50% lower in treated mice[1]. In mice, the growth of bovine tubercle bacilli in lung and spleen is inhibited by Kanamycin (2, 4 mg/kg; s.c.; 6 times a week for 3 weeks, once daily)[3]. Kanamycin (1.25, 5 mg/kg; s.c.; single dose at 3 h after infection) inhibits multiplication of K. pneumonia DT-S in the lung, trachea, and blood of mice, in proportion to the dose given, and also increases mouse survival.
