Scientific Background

PRAS40 (Proline-rich AKT1 substrate 1, AKT1S1) is a direct AKT substrate and an inhibitory component of mTORC1. AKT-mediated phosphorylation of PRAS40 at Thr246 relieves mTORC1 inhibition, linking PI3K/AKT signalling to downstream mTOR-dependent cell growth, protein synthesis, and autophagy. PRAS40 is a key node in oncogenic PI3K–AKT–mTOR signalling.

Recommended Protocol

Incubate the paired antibody (at working concentration) with a 5–10 molar excess of this peptide for 30–60 minutes at 4°C or room temperature. Centrifuge briefly if any precipitate forms, then use the pre-absorbed antibody in parallel with an unblocked control in the same experiment. Abolition or substantial reduction of signal confirms antibody specificity to the target epitope.
