Protein and DNA Removal Spin-Columns

What it means: Spin Column — the column format used to process each sample.

Why it matters:

• Spin columns need only a bench-top microcentrifuge and 1.5 ml tubes as receivers.
• Each column processes one sample, so several samples can be run in parallel in the same rotor.
• Separation takes a few short spins instead of a gravity column run.
• Pre-packed columns avoid packing resin by hand.
• The eluate is collected directly in the receiver tube; a final spin or 0.22 µm filtration removes any insoluble material.

For this product: Each spin column is pre-packed and ready to use after the preparation spins described in the ProFoldin protocol.

Tip: Cut the caps off the receiver tubes before use, as the protocol describes, so the column sits correctly during spins.

What it means: 20 columns — how many columns come in one pack.

Why it matters:

• Each column processes one 0.2 ml sample load, so one pack covers up to 20 samples.
• Larger sample volumes use more columns, or repeated loads on the same column where the protocol allows it.
• Including control samples, such as a buffer-only load, uses columns from the same pack.
• Columns are used one at a time, so unused columns can stay in the pack until needed.
• Running a whole sample series from one pack keeps the column preparation the same for every sample.

For this product: Each pack of this product contains 20 columns, each pre-packed and ready for the preparation spins.

Tip: Count samples, repeats and controls before starting so that one pack covers the whole experiment.

What it means: 0.2 ml — the sample volume loaded onto one column per spin.

Why it matters:

• Load 0.2 ml of sample in a buffer containing 100 mM NaCl; the salt helps keep small charged molecules off the column.
• Larger volumes are processed by repeating the load until the column binding capacity is reached.
• Staying within the stated volume keeps separation performance as described in the protocol.
• Loading the same volume on every column makes recovery easy to compare between samples.
• Have samples ready before starting the column preparation spins.

Tip: Measure 0.2 ml carefully for each load, and use the same volume for every column in a comparison.

What it means: More than 100 µg protein or 20 µg DNA — how much material one column can hold.

Why it matters:

• Proteins, DNA and RNA bind to the resin mainly through charge-charge interactions.
• Above the capacity, protein or nucleic acid can pass into the eluate.
• Organic and inorganic phosphates may also bind and use part of the capacity.
• The proteins, nucleic acids and any drug bound to them stay on the column, while small polar molecules pass into the eluate.

For this product: One column binds more than 100 µg protein or 20 µg DNA. Dilute or split samples that carry more than 100 µg of protein per 0.2 ml load; including 100 mM NaCl in the sample limits binding of small charged molecules.

Tip: Split concentrated samples across several columns to stay below the binding capacity.
