Overview

For quantitative determination of glutaminase activity in biological samples. The assay uses FL415/475 nm for signal readout. Compatible sample input includes Biological Samples, Purified Enzyme. Typical stated assay timing is 50 min.

Key elements and design rationale

  • Readout format: FL415/475 nm supports plate-based signal acquisition and consistent comparison across matched samples.
  • Sample compatibility: The stated sample scope includes Biological Samples, Purified Enzyme, which is useful when aligning matrix type with calibration and control design.
  • Analytical range context: The supplied specifications include a stated detection limit of 0.66 U/L for interpreting low-signal samples.
  • Feature emphasis: Fast and Safe. Assay can be completed within 40 minutes. Non-radioactive assay.

Additional feature notes highlight Sensitive and accurate. Linear detection range is 0.66-500 U/L glutaminase in a 96-well plate assay; Convenient and high-throughput. Homogeneous “mix-incubate-measure” type assay. Can be readily automated to assay thousands of samples per day. Available format information for this listing includes 100 Tests in 96-well plate.

Biological background

This product is centered on measurement of glutaminase within the matrices described for the assay. In practice, datasets from this type of format are typically interpreted by comparing relative signal, activity, or abundance across matched control and experimental groups rather than relying on a single value in isolation. Careful alignment of sample matrix, incubation window, and calibration strategy is important when comparing results across plates, operators, or study days.

More details

GLUTAMINASE(EC 3.5.1.2) is a key enzyme in the metabolism of glutamine. Glutamine is an important amino acid in the metabolism of cancer cells, and increased glutaminolysis has been linked to cancer. BioAssay Systems’ DGLN-100 Kit provides a convenient fluorimetric method to measure glutaminase activity in biological samples. In this assay, o-phthalaldehyde reacts with liberated ammonia where the increase in fluorescence at λex/em = 415/475 nm is directly proportional to enzyme activity.

Detection method

Fluorescent (FL 415/475 nm).

Reported detection limit: 0.66 U/L.

Procedures and timing

Stated procedure or timing information: 50 min.

Research relevance and current trends

  • Plate-based quantification and side-by-side group comparison remain central use cases for this assay format.
  • The product notes emphasize multi-sample throughput, making it relevant for screening-oriented and larger batch comparison studies.
  • Short assay timing and plate compatibility support time-course or repeated-measure collection plans when handling is kept consistent.

Common research applications

  • Quantify glutaminase in biological Samples, Purified Enzyme by FL415/475 nm readout.
  • Compare treatment or phenotype groups using matched biological Samples, Purified Enzyme handling.
  • Monitor time-course or pre/post changes in biological Samples, Purified Enzyme across study conditions.

Interpretation is usually strongest when signal changes are assessed alongside matrix-matched controls, replicate agreement, and the assay's stated analytical window.

Notes for experimental interpretation

  • Matrix composition, background signal, and sample handling can influence apparent response; compare like-with-like whenever possible.
  • Use appropriate blanks, controls, and replicate wells to distinguish biological differences from plate, reagent, or handling variability.
Can the kit?s sensitivity be increased?

Yes, you can increase the enzyme incubation step from 30 minutes to a time that provides detectable activity. The incubation time with the detection reagent will remain the same (20 minutes). Be sure to adjust the incubation time included in the calculation to represent the time you actually used.

Can I use this kit to determine the Km of my Glutaminase?

Yes, you can use this kit to determine the enzymatic parameters of your Glutaminase. The kit comes with a 20 mM Stock solution of the Glutamine substrate. You can use different dilutions of the substrate to generate the enzymatic parameters of your Glutaminase.

Does your kit include Glutaminase enzyme?

Our kit does not include enzyme. The customer needs to provide their enzyme of choice for this assay.
