Assay Principle
This sandwich ELISA quantifies rat 30 kDa adipocyte complement-related protein (gene Adipoq) in cell culture supernatants, serum, plasma (heparin, EDTA) and urine. Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
Adiponectin is a hormone secreted by adipocytes and one of the most abundant adipokines in plasma. Acting through the receptors ADIPOR1 and ADIPOR2, it activates AMPK and increases insulin sensitivity and fatty acid oxidation, and low circulating levels are associated with obesity and type 2 diabetes.
