Assay Principle
This sandwich ELISA quantifies rat angiopoietin-1 (gene Angpt1) in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). Strips are pre-coated with a capture antibody. After the sample is added, a biotinylated detection antibody binds the captured protein, and an avidin–biotin–peroxidase complex supplies the enzyme. TMB develops color in proportion to the bound target; the reaction is stopped and read at 450 nm, and concentrations are read from the standard curve.
Angiopoietin-1 (ANGPT1) is a secreted ligand that activates the endothelial receptor tyrosine kinase TIE2 (TEK). TIE2 signaling from ANGPT1 promotes endothelial survival and vessel maturation and keeps mature vessels stable and less leaky, so ANGPT1 is studied in angiogenesis, heart development and vascular disease.

Sample dilution: start at 1:100. In the supplier’s tests, serum and plasma samples measured about 120 ng/mL.
Example standard curve (OD450, pg/mL): 0 = 0.04; 156 = 0.109; 312 = 0.198; 625 = 0.311; 1250 = 0.685; 2500 = 1.131; 5000 = 1.69; 10000 = 2.289. Run a new standard curve on every plate.
Storage & Handling
Store at 4 °C for up to 6 months or −20 °C for up to 12 months. Avoid repeated freeze–thaw cycles. Ships on gel ice and tolerates up to 3 days at room temperature; freeze on arrival.
