Assay Principle
This single-incubation sandwich ELISA quantifies rat intercellular adhesion molecule 1 (gene ICAM1) in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). The strips are coated with an anti-tag antibody. Samples and standards are added to the wells together with the kit’s antibody cocktail (Antibody A and Antibody B); the resulting antibody–target complex is held on the plate through the tag. After washing, TMB substrate is added and the HRP label on the detection antibody converts it to a colored product in proportion to the bound target. The reaction is stopped and the plate is read at 450 nm. The supplier markets the Quick format as a 90-minute assay.
ICAM-1 (CD54) is a cell surface glycoprotein of the immunoglobulin superfamily that is strongly induced on endothelial and immune cells by inflammatory cytokines. It binds the integrins LFA-1 and Mac-1 on leukocytes, supporting their firm adhesion and passage across blood vessel walls as well as T-cell activation, and it is a widely used marker of endothelial activation and inflammation.
