Overview

Receptor tyrosine kinase binding ligands of the EGF family and activating several signaling cascades to convert extracellular cues into appropriate cellular responses. Known ligands include EGF, TGFA/TGF-alpha, amphiregulin, epigen/EPGN, BTC/betacellulin, epiregulin/EREG and HBEGF/heparin-binding EGF. Ligand binding triggers receptor homo- and/or heterodimerization and autophosphorylation on key cytoplasmic residues. The phosphorylated receptor recruits adapter proteins like GRB2 which in turn activates complex downstream signaling cascades. Activates at least 4 major downstream signaling cascades including the RAS-RAF-MEK-ERK, PI3 kinase-AKT, PLCgamma-PKC and STATs modules. May also activate the NF-kappa-B signaling cascade. Also directly phosphorylates other proteins like RGS16, activating its GTPase activity and probably coupling the EGF receptor signaling to the G protein-coupled receptor signaling. Also phosphorylates MUC1 and increases its interaction with SRC and CTNNB1/beta-catenin. Plays a role in enhancing learning and memory performance (By similarity). The epitope for this antibody is specifically expressed on EGFR-L858R mutated lung cancer.

This anti-EGFR antibody is supplied as Purified (Rabbit, Recombinant Rabbit Monoclonal, clone GFR/4564R, Rabbit IgG, Unconjugated) and is designed to support common target-detection workflows after the on-page specifications.

Key elements and design rationale

  • Target: EGFR
  • Format: Purified
  • Localization: Cytomembrane
  • Species reactivity: Human
  • Applications (listed): IHC-P
  • Conjugate: Unconjugated
  • Clone and antibody class: Recombinant Rabbit Monoclonal, clone GFR/4564R, Rabbit IgG

Because antibody performance can depend on epitope context, sample preparation, and biological state, interpret signals using appropriate controls and orthogonal evidence when possible.

Biological background

EGFR is referenced in public gene/protein resources (e.g., UniProt and NCBI Gene), which provide curated names/synonyms, protein features, and pathway context. When designing assays, consider potential isoforms, post-translational modifications, and cell-type specific expression that may influence observed signal.

Research relevance and current trends

  • Profiling EGFR expression across model systems, perturbations, and time points to support mechanistic hypotheses.
  • Combining antibody-based detection with multi-omics or imaging readouts to link EGFR signal with phenotype.
  • Using well-matched controls (isotype controls, genetic perturbations, or independent reagents) to strengthen interpretation of target-associated signal.

Common research applications

  • IHC-P

Use the listed applications as a starting point and tailor experimental design to your sample type and readout requirements.

Notes for experimental interpretation

  • Specificity considerations: closely related family members, isoforms, or PTMs can affect apparent specificity; confirm with independent approaches when critical.
  • Controls: include negative controls and, when feasible, genetic or pharmacologic perturbations to support target attribution in your system.
  • Species and sample context: differences in sequence, expression, fixation, or extraction conditions can change signal behavior across models.
