Product Overview
Recombinant Human EPO Protein (erythropoietin) is expressed in human cells and supplied as a glycosylated monomer. It is tag-free, animal-free and xeno-free, and is lyophilized from a 0.2 µm-filtered PBS solution without carrier protein, with purity >95% by SDS-PAGE and endotoxin <0.5 EU/µg.
EPO is used to support survival and maturation of EPOR-responsive erythroid progenitors, to study stage-specific erythroid commitment from CD34+ or pluripotent-cell-derived progenitors, to measure JAK2/STAT5, PI3K–AKT and MAPK responses, and in TF-1 proliferation and comparative potency assays. Expression in human cells supports mammalian glycosylation of the protein.
Expression system: Human cells · Tag-free · Animal-free
Scientific Background
Erythropoietin is a glycoprotein hormone made mainly by the kidney when oxygen is low. On erythroid progenitors it binds the erythropoietin receptor (EPOR); downstream of EPOR/JAK2, the STAT5, PI3K–AKT and MAPK pathways promote cell survival, proliferation and differentiation.
EPO acts mainly on committed erythroid progenitors, especially CFU-E and early erythroblasts, while SCF and other signals act at earlier stages. In the laboratory it is used to study erythropoiesis, erythroid progenitor biology, hemoglobinization and EPOR signaling.
Research Applications
EPO is used mainly at committed erythroid stages and in EPOR signaling and potency assays. The table shows typical placements; it is a planning guide, not a universal protocol.
| Application | Role of EPO | Companion context | Suggested readouts |
|---|---|---|---|
| CD34+ erythroid differentiation | Support of committed erythroid stages | SCF and stage-specific cytokine withdrawal | CD71, glycophorin A, hemoglobinization and enucleation |
| hPSC-derived erythropoiesis | Stage-specific erythroid maturation | Mesoderm and hematopoietic induction factors at earlier stages | CD43, CD45, CD235a, globin expression |
| EPOR signaling assay | Acute receptor activation | Serum-reduced conditions and time course | pJAK2, pSTAT5, pAKT and target genes |
| TF-1 potency assay | Dose-dependent proliferation | Defined assay medium | Proliferation, viability and relative potency |
Planning considerations
- Define the responsive cell stage and confirm EPOR expression or pathway competence.
- Begin with conditions from a validated protocol and titrate EPO in the actual cells, medium and endpoint; no numerical EC50 or universal working concentration is given for this product.
- Control timing and companion factors such as SCF, IL-3 and TPO, since signal order and duration can change lineage or functional outcomes.
- Include no-factor, vehicle and pathway-response controls, and measure phenotype and function rather than cell number alone.

Biological Activity & Quality Control
Biological activity is measured by stimulation of human TF-1 cell proliferation, with a concentration-dependent response.
Lot-specific activity and purity data are provided on the Certificate of Analysis (CoA), available from BioHippo on request.
Reconstitution
- Briefly centrifuge the vial before opening.
- Reconstitute in sterile PBS containing 0.1% endotoxin-free recombinant human serum albumin.
- Mix gently and prepare aliquots.
