Product Overview
Recombinant Human FLT3 Ligand Protein (FLT3L) is a hematopoietic cytokine expressed in human cells and supplied as a glycosylated monomer. It is tag-free, animal-free and xeno-free, and is lyophilized from a 0.2 µm-filtered PBS solution without carrier protein, with purity >95% by SDS-PAGE and endotoxin <0.5 EU/µg.
FLT3 Ligand is used in hematopoietic stem/progenitor-cell culture, usually together with SCF and TPO, in dendritic-cell development and in staged differentiation of immune cells from pluripotent stem cells, as well as in FLT3/CD135 signaling studies. Expression in human cells supports mammalian folding and glycosylation, and five peer-reviewed studies identify this FLT3 Ligand or its catalog number.

Scientific Background
FLT3 Ligand is a glycoprotein cytokine produced by several stromal and immune-cell populations. The soluble form is released from a membrane-associated precursor and binds the receptor tyrosine kinase FLT3/CD135 on early hematopoietic progenitors.
Receptor activation engages PI3K–AKT, RAS–MAPK and STAT signaling, and the result depends on progenitor state and companion cytokines. FLT3L acts most clearly on early progenitors and dendritic-cell lineages, and is studied with SCF and TPO in HSPC culture and with stage-specific signals in dendritic, lymphoid and other immune-cell differentiation.
Research Applications
FLT3 Ligand is mostly used in combination with other cytokines to support FLT3-positive early progenitors and dendritic-cell lineages. The table shows typical placements; it is a planning guide, not a universal protocol.
| Application | Role of FLT3 Ligand | Companion context | Suggested readouts |
|---|---|---|---|
| Cord-blood or adult CD34+ HSPC culture | Survival and expansion of early progenitors | SCF and TPO, plus protocol-dependent signals | Cell count, viability, CD34/CD38/CD90, CFU and engraftment assays |
| hPSC hematopoietic differentiation | Support after hemogenic/endothelial stages | SCF, TPO, IL-3, IL-7 and/or Notch context | CD43, CD45, CD34, lineage markers and functional assays |
| Dendritic-cell research | Expansion of FLT3-responsive progenitors | Lineage-specific maturation signals | DC subset markers, antigen presentation and cytokine output |
| FLT3 signaling assay | Receptor stimulation | Defined serum-reduced or basal system | Phospho-FLT3, ERK, AKT, STAT5 and proliferation |
Planning considerations
- Define the developmental or immune-cell stage and confirm that the target population expresses FLT3/CD135 or depends on the pathway.
- Take the starting dose from published or in-house qualified work and titrate it for each cell source, basal medium and readout; no universal working concentration or EC50 applies across systems.
- Control timing and companion cytokines, since signal order, duration and pathway interactions shape the outcome.
- Include no-factor, vehicle and pathway-response controls, and assess identity, viability, signaling and function rather than cell number alone.
