Product Overview
Recombinant Human GM-CSF Protein is a full-length, glycosylated monomer expressed in human cells. It is manufactured in an animal-free, xeno-free and tag-free format and supplied lyophilized without carrier protein, at >95% purity by SDS-PAGE and endotoxin <0.5 EU/µg.
GM-CSF is widely used with IL-4 to generate immature monocyte-derived dendritic cells, and in studies of myeloid progenitor survival, proliferation and differentiation, antigen presentation, cytokine production and GM-CSF receptor signaling. Expression in human cells supports mammalian folding and glycosylation. Activity is confirmed by TF-1 cell proliferation.
Expression system: Human cells · Tag-free · Animal-free
Scientific Background
GM-CSF (CSF2) is a four-helix-bundle cytokine produced by activated T cells, macrophages, endothelial cells and stromal cells. Binding to its receptor complex triggers JAK2/STAT5, MAPK and PI3K signaling, which supports survival, proliferation and functional maturation of myeloid progenitors and mature myeloid cells.
In vitro, GM-CSF is a standard factor for monocyte-derived dendritic-cell generation, often together with IL-4, and for studying granulocyte and macrophage lineage responses. Results depend on cell source, cytokine dose, exposure time and companion signals.
Research Applications
GM-CSF supports responsive myeloid populations, from progenitors to monocyte-derived dendritic cells. The table summarizes where it is typically placed in a workflow; it is a planning guide, not a universal protocol.
| Application | Role of GM-CSF | Companion signals and context | Suggested readouts |
|---|---|---|---|
| Monocyte-derived dendritic cells | Supports early differentiation | IL-4; later a maturation cocktail | CD11c, HLA-DR, CD80/CD86, morphology and antigen presentation |
| Myeloid progenitor culture | Survival and lineage support | SCF, IL-3 or lineage-specific factors | Cell count, viability, CFU, CD14/CD15/CD33 |
| TF-1 bioassay | Receptor-dependent proliferation | Defined low-serum assay conditions | Dose response, viability and proliferation |
| Myeloid signaling | Acute receptor activation | Time-course and pathway controls | pSTAT5, pERK, pAKT and target genes |
Planning considerations
- Define the responsive cell stage and confirm that the cells express the GM-CSF receptor.
- Begin with a validated protocol, then run a dose response for the actual cell source, medium and endpoint.
- Control timing and companion factors such as IL-4, since signal order and duration can change lineage or functional outcomes; include no-factor, vehicle and pathway-response controls.
- Measure phenotype and function rather than cell number alone. No EC50 or universal working concentration applies without lot- or protocol-specific evidence.
