Scientific Background
Interleukin-1 Receptor Antagonist (IL-1RN) is a member of the IL-1 family. Endogenous IL-1RN is produced in numerous animal disease models as well as in human autoimmune and chronic inflammatory diseases. It binds to IL-1 receptors in competition with IL-1, but does not elicit intracellular response from this binding. Its role in counteracting the proinflammatory effects of IL-1 is being studied by numerous research groups. IL-4 and IL-13 have been shown to amplify the stimulatory effect of IL1-beta on the production of soluble and intracellular forms of IL-1RN. The regulated expression of IL-1RN in various cell types has been shown to be influenced by cytokines.

This protein was produced in E. coli expression system. The expression system determines the protein post-translational modification profile, folding characteristics, and suitability for specific applications. Mammalian (HEK293) expression is used where native-like glycosylation is required; insect cell and E. coli systems are used for high-yield non-glycosylated production.
This product is supplied as lyophilized powder. Centrifuge the vial briefly before opening, then add the recommended volume of sterile water or the specified buffer and allow to dissolve at 4°C for 30 minutes without vortexing. Prepare single-use working aliquots in 0.1% BSA to minimise freeze-thaw losses of the bulk stock.
Endotoxin is < 1.0 EU per μg of the protein as determined by the LAL method., determined by the LAL chromogenic method. The <1.0 EU/µg specification ensures suitability for cell culture experiments, primary cell stimulation, and assays measuring immune activation or cytokine secretion, where endotoxin interference with NF-κB signalling is a concern.
This protein meets > 95% as determined by reducing SDS-PAGE purity as assessed by reducing SDS-PAGE. A gel image for each lot is included on the Certificate of Analysis. Contact technical support if purity >98% is required for biophysical measurements such as crystallography, ITC, or single-particle cryo-EM.
Generally, lyophilized proteins are stable for up to 12 months when stored at -20 to -80℃. Reconstituted protein solution can be stored at 4-8℃ for 2-7 days. Aliquots of reconstituted samples are stable at < -20℃ for 3 months. Shelf life is 12 months from the date of receipt. Prepare small working aliquots to minimise freeze-thaw cycles of the bulk stock. Reconstituted lyophilized protein is stable for up to 3 months at −80°C in carrier protein.
