Product Overview
Recombinant Human IL-2 Protein is a full-length, glycosylated monomer expressed in human cells. It is manufactured in an animal-free, xeno-free and tag-free format and supplied lyophilized without carrier protein, at >95% purity by SDS-PAGE and endotoxin <0.5 EU/µg.
IL-2 is used to support proliferation and survival of activated T cells, to expand NK cells for functional and cytotoxicity assays, to expand Vδ2+ and other γδ T-cell populations, and to study IL-2 receptor, STAT5 and survival signaling. Activity is confirmed by CTLL-2 cell proliferation, and two peer-reviewed studies identify this IL-2 or its catalog number.

Scientific Background
Interleukin-2 is a glycosylated, short-range cytokine produced mainly by activated T cells. It drives proliferation and survival of activated T cells, can stimulate NK cells and also contributes to the maintenance of regulatory T cells.
IL-2 signals through IL-2 receptor complexes and activates JAK1/JAK3–STAT5 together with PI3K–AKT and MAPK pathways. Receptor composition and IL-2 concentration strongly shape which cells respond, and outcomes also depend on activation state, culture density, companion cytokines such as IL-7, IL-15 and IL-21, and exposure time.
Research Applications
IL-2 supports the expansion and survival of several lymphocyte populations and is a standard ligand for IL-2 receptor signaling studies. The table summarizes typical workflow placement; it is a planning guide, not a universal protocol.
| Application | Role of IL-2 | Companion signals and context | Suggested readouts |
|---|---|---|---|
| Activated conventional T cells | Proliferation and survival | TCR/CD3-CD28 or antigen activation | Expansion, viability, phenotype, cytokines and cytotoxicity |
| NK-cell culture | Growth and activation support | IL-15 and/or IL-21 depending on protocol | CD56/CD16 phenotype, degranulation, killing and cytokines |
| γδ T-cell culture | Expansion after phosphoantigen-related activation | Zoledronate/pamidronate or IPP in protocol-specific systems | Vδ2 frequency, proliferation, cytokines and tumor-cell killing |
| Regulatory T-cell research | Context-dependent survival and STAT5 support | Defined TCR stimulation and low-dose experimental design | FOXP3, CD25, suppressive function and pSTAT5 |
Planning considerations
- Define the immune-cell stage and confirm that the target population expresses the relevant IL-2 receptor components.
- Start from a peer-reviewed or internally qualified concentration, then run a dose response for the cell source, basal medium and readout.
- Control timing and companion cytokines, since effects depend on signal order, duration and pathway interactions; include no-factor, vehicle and pathway-response controls.
- Assess identity, viability, signaling and function rather than cell number alone. No EC50 or universal working concentration applies without lot- or protocol-specific evidence.

Biological Activity & Quality Control
Activity is determined in a CTLL-2 assay, in which the protein drives proliferation of these mouse cytotoxic T cells in a dose-dependent way.
Lot-specific activity and purity data are provided on the Certificate of Analysis (CoA), available from BioHippo on request.
Reconstitution
- Briefly centrifuge the vial before opening.
- Reconstitute in sterile PBS containing 0.1% endotoxin-free recombinant human serum albumin.
- Mix gently and prepare single-use aliquots under sterile conditions.
- Avoid repeated freeze–thaw cycles; a manual-defrost freezer is recommended.
Storage & Shipping
| Storage | As supplied: −20°C to −80°C for up to 12 months from receipt. After reconstitution: 2°C to 8°C for up to 1 month, or −20°C to −80°C for up to 3 months, under sterile conditions. Avoid repeated freeze–thaw cycles. |
