Scientific Background
Human IL-36 Receptor Antagonist Protein/IL-36RN/IL-1F5 — also known as FIL; UNQ; PRO; IL1F; IL1L; IL1HY; IL1RP; PSORS; IL36RN; FIL1; FIL1(DELTA); FIL1D; IL-36Ra; IL1F5; IL1HY1; IL1L1; IL1RP3; — Human Interleukin-36 Receptor Antagonist (IL-36RN) is a secreted protein which belongs to the Interleukin 1 cytokine family (IL-1 family). IL-36RN is predominantly expressed in keratinocytes but not in fibroblasts, endothelial cells or melanocytes. IL-36RN is also detected in the spleen, brain leukocyte and macrophage cell types. Increased in lesional psoriasis skin. IL-36RN is a highly and a specific antagonist of the IL-1 receptor-related protein 2-mediated response to Interleukin 1 family member 9 (IL1F9). Dysregulated expression of novel agonistic and antagonistic IL-1 family member ligand
Research into Human IL-36 Receptor Antagonist Protein/IL-36RN/IL-1F5 supports investigations in immune regulation and inflammatory responses, with implications for cytokine biology.

Biological Activity
Activity ValidatedMeasured by its ability to inhibit IL-36 alpha, IL-36 beta or IL-36 gamma-induced IL-8 secretion in A431 human epithelial carcinoma cells. The ED50 for this effect is 23.14 ng/ml.
Perform a titration in your specific assay format to establish optimal working concentration. Include a previously validated lot as an in-house reference when qualifying new lots.

This protein is produced in E. coli (prokaryotic). E. coli expression gives high yield without glycosylation — verify that your assay tolerates non-glycosylated protein.
Purity: > 95% as determined by reducing SDS-PAGE., confirmed by reducing SDS-PAGE. Endotoxin level: < 1.0 EU per μg of the protein as determined by the LAL method., measured by LAL assay — critical for cell-based studies. Identity is verified by peptide mapping or N-terminal sequencing on request.
Yes — biological activity is confirmed per lot. Activity data: Measured by its ability to inhibit IL-36 alpha, IL-36 beta or IL-36 gamma-induced IL-8 secretion in A431 human epithelial carcinoma cells. The ED50 for this effect is 23.14 ng/ml.. Run a titration in your own system to establish optimal working concentration before committing all samples.
Reconstitution: Please refer to the printed manual for detailed information.. Storage: Generally, lyophilized proteins are stable for up to 12 months when stored at -20 to -80℃. Reconstituted protein solution can be stored at 4-8℃ for 2-. Avoid repeated freeze-thaw — each cycle risks partial denaturation.
This protein is validated for ELISA, Functional Assay (In Vitro). It is commonly used as an ELISA standard, binding partner in SPR/BLI, or stimulant in cell-based functional assays. Validate performance in your specific format before use in critical experiments.
