Product Overview
Recombinant Human Noggin Protein is a full-length, glycosylated monomer expressed in human cells. Noggin is a secreted antagonist of bone morphogenetic proteins (BMPs). The protein is made in an animal-free, xeno-free and tag-free format and supplied lyophilized without carrier protein, at >95% purity by SDS-PAGE and endotoxin <0.5 EU/µg.
Researchers use Noggin to block extracellular BMP signaling during neural induction and neural progenitor work, to keep a low-BMP environment in intestinal, gastric, neural and other organoid systems, and to study BMP-dependent patterning of neural, mesodermal and skeletal tissues. It also serves as a defined antagonist for titrating BMP-2/BMP-4 responses in signaling assays. Function is confirmed in a cell-based BMP-4 antagonism assay.
Expression system: Human cells · Tag-free · Animal-free
Scientific Background
Noggin is a secreted cystine-knot protein that binds selected BMPs, most notably BMP-2 and BMP-4, and prevents them from reaching their type I and type II receptors. Less BMP–receptor engagement means less SMAD1/5/8 activation, which shifts cell-fate decisions in a way that depends strongly on developmental stage and context.
In vivo, Noggin has key roles in neural patterning, skeletal development and the formation of cartilage and joints. In culture it is not a universal differentiation factor: the outcome depends on the endogenous BMP level, Noggin concentration, matrix, cell state and companion pathways such as FGF, EGF, WNT and Activin/Nodal.
Research Applications
Noggin is used wherever BMP signaling needs to be reduced at a defined stage, from neural induction to organoid expansion. The table shows where it is typically placed in a workflow; it is a planning guide, not a universal protocol.
| Application | Role of Noggin | Companion signals and context | Suggested readouts |
|---|---|---|---|
| Neural induction | Lowers BMP-dependent signals toward non-neural fates | Often combined with TGF-β/Activin/Nodal inhibition in dual-SMAD protocols | PAX6, SOX1, NESTIN, neural rosettes and lineage competence |
| Organoid expansion | Keeps a tissue-specific low-BMP niche | EGF, FGF, WNT/R-spondin or tissue-specific factors | Organoid-forming efficiency, morphology, lineage markers and function |
| BMP signaling assay | Extracellular antagonist | BMP-2 or BMP-4 dose-response control | pSMAD1/5/8, ID genes, alkaline phosphatase and reporter activity |
| Skeletal/chondrogenic research | Modulates BMP-driven differentiation | Matrix and stage-specific chondrogenic signals | ALP, SOX9, COL2A1, mineralization and morphology |
Planning considerations
- Define the developmental stage and confirm that the target cells depend on BMP signaling at that stage.
- Start from a validated or internally qualified concentration, then run a dose response for the cell source, basal medium and readout.
- Control timing and companion factors, since signal order and duration change the outcome; include no-factor, vehicle and BMP-response controls.
- Assess identity, viability, signaling and function rather than cell number alone. No universal working concentration or EC50 applies without lot- or protocol-specific evidence.

Biological Activity & Quality Control
Biological activity is measured by the ability of Noggin to inhibit BMP-4-induced alkaline phosphatase production in ATDC-5 mouse chondrogenic cells.
Lot-specific activity and purity data are provided on the Certificate of Analysis (CoA), available from BioHippo on request.
Reconstitution
- Briefly centrifuge the vial before opening.
- Reconstitute in sterile PBS containing 0.1% endotoxin-free recombinant human serum albumin.
- Mix gently and prepare single-use aliquots under sterile conditions.
- Avoid repeated freeze–thaw cycles; a manual-defrost freezer is recommended.
