Kex2 protease, also known as a double-alkaline endopeptidase or YSCF protease, is a calcium-dependent serine protease that is responsible for processing the precursors of killer toxin and α-factor within yeast cells. It specifically recognizes and cleaves the peptide bonds at the carboxyl end of dibasic amino acids such as Arg-Arg and Lys-Arg. Unlike trypsin, Kex2 does not recognize and cleave the peptide bonds at the carboxyl end of single basic amino acids, such as arginine or lysine. The activity of Kex2 protease is not inhibited by conventional serine protease inhibitors such as PMSF, TPCK.
YeasenUCF.METM Recombinant Kex2 Protease is expressed in Pichia pastoris, produced under GMP regulations, free of any animal-derived components, and without the risk of viral contamination from animal sources. It has the same enzymatic specificity as the natural Saccharomyces cerevisiae Kex2 enzyme. The optimal pH for activity is pH 9.0, and the stable storage pH is between 5.0 and 6.0.

Shipping and Storage
Lyophilized powder can be stored at 2~8℃ for one year, protected from light and moisture.
Dissolution and Storage Buffer (20 mM NaAc-HAc, pH 5.2, 2 mM Ca2+), after dissolution, store at below -20℃, it is recommended to aliquot for storage to avoid repeated freezing and thawing.

Store this enzyme at 2~8°C and avoid repeated freeze-thaw cycles to preserve catalytic activity. The product is shipped Gel Pack and remains stable for up to one year from the date of manufacture when stored under recommended conditions. Aliquoting the stock solution into single-use volumes is recommended for enzymes used infrequently to minimize thermal cycling of the bulk stock.

One unit (U/mg) is defined as the amount of protease activity per milligram of enzyme protein under standardized assay conditions (substrate concentration, pH, temperature, and incubation time).
This enzyme is produced as Recombinant (E. coli) and is available in GMP grade for regulated applications. Each lot is subjected to activity assay, purity assessment by SDS-PAGE, and functional validation prior to release. Most serine proteases (e.g., Proteinase K) are inhibited by PMSF (1 mM) and heat-inactivated at 95°C for 10 min. Proteinase K retains activity across a broad pH range (4–12) and tolerates SDS, urea, and other denaturants, making it particularly effective for nucleic acid extraction from challenging samples including FFPE tissue.
