Compound Overview

RGB-1 selectively binds and stabilizes RNA G-quadruplex structures, thereby reducing expression of the NRAS proto-oncogene in breast cancer cells. It can serve as a tool for investigating the cellular functions of RNA G-quadruplex structures and identifying novel mRNA G-quadruplex-forming sequences, and it is applicable to breast cancer research[1]. It is a light yellow to yellow solid (C22H12N4O4S2, MW 460.49).

In Vitro

RGB-1 binds specifically to the TERRA RNA G-quadruplex with an apparent Kd of 5.9 μM and shows minimal to no binding to DNA G-quadruplex structures[1]. In a cell-free system, RGB-1 selectively reduces translation of mRNA that carries a TERRA RNA G-quadruplex in its 5' UTR[1]. RGB-1 (5-20 μM) reduces NRAS protein expression in MCF-7 cells in a dose-dependent manner, without changing NRAS mRNA levels[1].
