Compound Overview

RIPK3-IN-1 is a RIPK3 type II DFG-out inhibitor with an IC50 of 9.1 nM. It also inhibits RIPK1 and RIPK2, with IC50 values of 5.5 μM and >10 μM, respectively, and acts as a c-Met kinase inhibitor with an IC50 of 1.1 μM[1]. It is supplied as a light yellow to yellow solid (C29H25FN4O4, MW 512.53) at 99.30% purity.

For Research Use Only. Not for use in diagnostic or therapeutic procedures, and not for human or veterinary use. Handle in accordance with the Safety Data Sheet and your institution's chemical hygiene plan.

In Vitro

SolventSolubilityNotes
DMSO5 mg/mL (9.76 mM)requires sonication and warming and heat to 80°C; use freshly opened DMSO (absorbed moisture lowers solubility)

Aliquot the stock solution and store it at -80°C (up to 2 years) or -20°C (up to 1 year); avoid repeated freeze-thaw cycles.

In Vitro

ABL, BRAF/V599E, MAP4K3, and SRC are also inhibited by RIPK3-IN-1, with IC50s of 0.37, 0.15, 0.012, and 0.075 μM, respectively[1]. Necroptosis, a programmed form of cell death, has been linked to many diseases, among them ischemia reperfusion injury, neurodegenerative disorders, pancreatic cancer, and autoimmune conditions such as inflammatory bowel disease (IBD). Stimulation of death receptors (for example, the TNF receptor family) can trigger signaling that starts necroptotic cell death. The process involves a necrosome, a cytosolic complex containing receptor interacting protein kinases 1 and 3 (RIPK1, RIPK3)[1].
