Compound Overview

RNA polymerase II-IN-2 inhibits RNA polymerase II, with a Ki of 74.1 nM, and it inhibits Pol II-mediated transcription while inducing cytotoxicity in mammalian cells. It serves as a payload for antibody-drug conjugates (ADCs) and has been applied in cancer research[1]. It has the molecular formula C41H58N10O12S (MW 915.02).

In Vitro

RNA polymerase II-IN-2 (20iii) is nearly twice as cytotoxic as α-amanitin to CHO cells (normalized IC50 value of 0.61)[1]. RNA polymerase II-IN-2 is 5-fold more cytotoxic than α-amanitin in HEK293 cells (normalized IC50 value of 0.20)[1]. RNA polymerase II-IN-2 shows greater cytotoxicity than α-amanitin toward HeLa cells, with a normalized IC50 value of 0.60[1]. Toward HepG2 liver-derived cells, RNA polymerase II-IN-2 is significantly less cytotoxic than α-amanitin, with a normalized IC50 value of 2.7[1].
