Product Overview

RNase Inhibitor is a recombinant RNase inhibitor expressed in soluble form in Escherichia coli. It performs equivalently to the ribonuclease inhibitor purified from human placenta. It is a protein of approximately 51 kDa with an isoelectric point (pI) of 4.7. RNase Inhibitor can specifically bind RNase A, B, and C with a non-covalent bond to form a 1:1 complex to inactivate RNase, and has a broad spectrum of RNase inhibitory activity. The inhibitor is active in buffers of 0–0.5 M NaCl, pH 5–8, and has the highest activity at pH 7.8. It protects the integrity of mRNA and improves the efficiency of transcription and translation, while avoiding the possible effects of using organic compound inhibitors. The inhibitor is compatible with commonly used reverse transcriptases and DNA polymerases in RT-PCR and RT-qPCR. Unlike the human placental inhibitor, the recombinant protein lacks two cysteine residues and is therefore more resistant to oxidation, making it better suited to reactions that are sensitive to high DTT, such as qPCR.

The inhibitor is supplied as a single-component solution at 40 U/µL in 20 mM HEPES-KOH (pH 7.5), 50 mM KCl, 5 mM DTT and 50% glycerol. One activity unit is the amount of enzyme that inhibits 50% of the activity of 5 ng of RNase A. SDS-PAGE shows a single band at a molecular weight of 50 kDa.
