The RT-LAMP Fluorescent Assay Kit (Bst Plus) is a kit capable of isothermal amplification using DNA and RNA as templates. This kit employs the next-generation aptamer-encapsulated Bst Plus DNA Polymerase and heat-resistant HifairTM III Reverse Transcriptase, along with an MRI, combined with a finely optimized buffer system, to achieve highly efficient, rapid, and highly specific detection reactions, and supports room-temperature reaction system preparation.

Specificity was evaluated using both DNA and RNA templates, including positive targets and non-target (negative) controls. The RT-LAMP assay (Cat#16729) showed no false positives and clear separation between positive and negative samples, outperforming Supplier A* by avoiding non-specific amplification.

RT-LAMP and LAMP reactions in this kit proceed isothermally at 60–65°C (optimized temperature specified in the datasheet). A standard heat block, water bath, or isothermal dry-bath incubator is sufficient — no thermocycler is required. This makes the kit well-suited to resource-limited or point-of-care settings where conventional PCR instrumentation is unavailable.
The Hotstart Bst Plus enzyme used in this kit incorporates an antibody-mediated cold-block that prevents non-specific amplification and false-positive results at ambient temperature. Activity is released upon incubation at ≥60°C, improving assay specificity and enabling pre-reaction setup at room temperature without risk of non-specific product formation.
Detection method depends on the kit format: Dye Assay kits use intercalating fluorescent dyes (e.g., SYBR Green, calcein, or YSFluor) for visual or instrument-based detection; Fluorescent Assay kits use real-time fluorescence monitoring; Probe Assay kits use sequence-specific probes for enhanced specificity. This kit is validated for purified RNA or DNA templates. Some formats support direct amplification from swabs, saliva, or crude lysates without prior nucleic acid extraction — check the specific product datasheet for the validated sample input types, inhibitor tolerances, and any pre-treatment requirements.
LAMP amplification typically completes within 20–60 minutes at the isothermal incubation temperature, compared to 1.5–3 hours for conventional RT-PCR. The faster time-to-result makes this kit especially useful for high-throughput screening workflows and time-sensitive applications such as outbreak response and quarantine protocols.
