Bacterial DNA topoisomerase II (gyrase) is a type II topoisomerase that converts relaxed circular DNA into negatively supercoiled DNA. Type II topoisomerases cut and rejoin both DNA strands to change DNA topology.
Some inhibitors, such as fluoroquinolones, block the rejoining step; the enzyme then stays covalently linked to cut DNA in a cleavage complex. Ciprofloxacin and novobiocin both inhibit gyrase, but only ciprofloxacin produces cleavage complexes.
This product is the GyrA subunit of S. aureus DNA gyrase. DNA gyrase is active as a heterotetramer of two subunit types, so GyrA alone has no supercoiling activity; combine it with GyrB (S. aureus GyrB, BHP16300037) to reconstitute the active enzyme. The DNA gyrase holoenzyme is also sold separately as S. aureus DNA Gyrase, BHP16300035.
