Scientific Background
Staphylococcus aureus is a leading cause of community- and hospital-acquired infections worldwide, including bacteremia, endocarditis, and device-related infections. Approximately 80% of bacteremic S. aureus isolates express capsular polysaccharide of serotype 5 (CP5) or serotype 8 (CP8). CP5 is a linear polysaccharide composed of trisaccharide repeating units and serves as a virulence factor by shielding the bacterium from opsonophagocytic killing. As the dominant serotype in many clinical bloodstream isolates, CP5 is a key vaccine antigen candidate and a critical reference reagent for opsonophagocytic activity (OPA) assays and ELISA-based serology.

CP5 is used as a reference antigen in opsonophagocytic activity (OPA) assays to evaluate the functional efficacy of anti-S. aureus antibodies. It is also used for ELISA development, serology, passive immunization studies, and as a vaccine antigen in S. aureus vaccine research programs.

CP5 (serotype 5) and CP8 (serotype 8) are structurally distinct capsular polysaccharides from S. aureus, differing primarily in the configuration of the N-acetyl-fucosamine residues. Together they account for capsular polysaccharide expression in approximately 80% of bacteremic clinical isolates. When developing serology assays or evaluating vaccine coverage, including both serotypes is recommended.
Store at -20°C for short-term use. For long-term archival storage, keep at -80°C. Avoid repeated freeze-thaw cycles. Upon receipt, aliquot into single-use volumes to maximize stability.
