Scientific Background

Staphylococcus aureus is a leading cause of community- and hospital-acquired infections worldwide. Together, capsular polysaccharide serotypes 5 (CP5) and 8 (CP8) account for surface polysaccharide expression in approximately 80% of clinical bloodstream isolates. CP8 is a linear polysaccharide with trisaccharide repeating units that differ from CP5 in the anomeric configuration of the N-acetyl-fucosamine residue. Serotype 8 strains, including Reynolds and O13, are prevalent among nosocomial and community-acquired bacteremia cases. CP8 is a clinically important vaccine antigen candidate and a reference reagent for serotype 8-specific OPA assays and anti-S. aureus antibody quantification.

What is S. aureus Purified CP8 used for?

CP8 is used as a reference antigen in opsonophagocytic activity (OPA) assays, ELISA development, and serology for evaluating anti-S. aureus immune responses. It is also used as a vaccine antigen in S. aureus vaccine research.

How does CP8 differ from CP5?

CP5 and CP8 are structurally related but serologically distinct capsular polysaccharides from S. aureus. They differ in the anomeric configuration of the N-acetyl-fucosamine residues (C4-linked in CP5 vs. C3-linked in CP8). CP5 has a Kav of 0.038 while CP8 has a Kav of 0.026, reflecting their different molecular weight profiles. Antibodies against one serotype do not cross-react with the other, making it essential to include both serotypes in comprehensive S. aureus serology panels.

What strains was CP8 purified from?

This CP8 product is purified from S. aureus strains Reynolds and O13 — two reference strains that are widely used in S. aureus serotype 8 research and OPA assay development. Quality is confirmed using the CP8-specific monoclonal antibody 5A6.

How should CP8 be stored?

Store at -20°C for short-term use. For long-term storage, keep at -80°C. Avoid repeated freeze-thaw cycles. Aliquot into single-use volumes upon receipt to preserve product integrity.
