- Also known as: Fluoro-willardiine, (S)-(−)-α-Amino-5-fluoro-3,4-dihydro-2,4-dioxo-1(2H)-pyrimidinepropanoic acid
- CAS number: 140187-23-1
- Molecular formula: C7H8FN3O4.
- Purity: >97%
- Concentration: 10 nM - 1 µM.
- Form: Lyophilized Powder
- Solubility: Soluble up to 50 mM in 1.1eq NaOH, with gentle heating. Centrifuge all products before handling (10000 x g 5 min).
- Reconstitution: Centrifuge the vial before adding solvent (10,000 x g for 5 minutes) to spin down all the powder to the bottom of the vial. The lyophilized product may be difficult to visualize. Add solvent directly to the centrifuged vial. Tap the vial to aid in dissolving the lyophilized product. Tilt and gently roll the liquid over the walls of the vial. Avoid vigorous vortexing. Light vortexing for up to 3 seconds is acceptable if needed. For long-term storage in solution, we recommend preparing a stock solution by dissolving the product in sterile water at a concentration of at least 0.1 mg/mL. Divide the stock solution into small aliquots and store at -20°C. Before use, thaw the relevant vial(s) and dilute to the desired working concentration in your working buffer. It is recommended to prepare fresh solutions in working buffers just before use. For long-term storage of diluted solutions, we recommend adding 0.1% BSA. Repeat freeze-thawing may result in loss of activity.
- Shipping: Shipped at room temperature. Product as supplied can be stored intact at room temperature for several weeks. For longer periods, it should be stored at -20°C.
- Target: AMPA receptors
- Source: Synthetic
- Activity: (S)-(-)-5-Fluorowillardiine is a potent and selective AMPA receptor agonist (Ki values of 14.7 nM and 25.1 nM, on hGluR1 and hGluR2, respectively), selective over kainate receptors (Ki of 1.82 µM on hGluR5). More potent and selective than AMPA1-2.
- Alternative names: Fluoro-willardiine, (S)-(−)-α-Amino-5-fluoro-3,4-dihydro-2,4-dioxo-1(2H)-pyrimidinepropanoic acid
Scientific background
(S)-(-)-5-Fluorowillardiine is a highly potent and selective AMPA receptor agonist. 5-Fluorowillardiine is synthesized as two distinct isomers: R and S. (S)-(-)-5-Fluorowillardiine is selective over kainate receptors and is more potent and selective than AMPA itself; at equilibrium, (S)-(-)-5-Fluorowillardiine is seven times more potent than AMPA and 30 times more potent than willardiine. (S)-(-)-5-Fluorowillardiine demonstrates an EC50 value of 1.5 µM1,2.The ionotropic glutamate AMPA receptors (AMPARs) are the primary receptors mediating fast excitatory synaptic transmission in the mammalian brain. This function is essential for synaptic plasticity and learning and memory3,4.
