SB203580 is a pyridinyl imidazole compound that selectively inhibits p38 MAP kinase, particularly the p38α isoform. It also inhibits phosphorylation of heat shock protein 27 (HSP27) and can activate Raf-1 signaling. In neuroscience, SB203580 is used to study the role of p38 MAPK in neuroinflammation, synaptic plasticity, and neuronal survival. Its high selectivity over kinases such as LCK, GSK-3β, and PKBα makes it a reliable tool for dissecting p38-mediated signaling in neurodegenerative disease models. SB203580 is cell-permeable and is frequently employed in studies of Alzheimer's disease, stroke, and other CNS disorders involving inflammatory and stress responses.
