In Vivo

Choose the formulation that suits the animal model and route of administration; percentages are volume ratios of the final working solution. Start from a clear DMSO stock (see In Vitro above), add the co-solvents one at a time in the order listed, mixing after each addition, and prepare the working solution fresh on the day of dosing. If precipitation or phase separation occurs, gentle warming or sonication can help.

How to prepareGives a suspension at 5 mg/mL. The suspension is suitable for oral and intraperitoneal dosing. For 1 mL of working solution: add 100 μL DMSO stock (50.0 mg/mL) to 400 μL PEG300; then 50 μL Tween-80; then 450 μL saline to bring the volume to 1 mL. Saline: dissolve 0.9 g sodium chloride in ddH2O and make up to 100 mL.

How to prepareGives a suspension at 5 mg/mL. The suspension is suitable for oral and intraperitoneal dosing. For 1 mL of working solution: add 100 μL DMSO stock (50.0 mg/mL) to 900 μL 20% SBE-β-CD in saline. 20% SBE-β-CD in saline: dissolve 2 g SBE-β-CD powder in 10 mL saline until clear (4°C, store up to one week).

How to prepareGives a clear solution at ≥ 5 mg/mL (saturation not determined). Use with caution if continuous dosing will exceed two weeks. For 1 mL of working solution: add 100 μL DMSO stock (50.0 mg/mL) to 900 μL corn oil.

In Vitro

Seco Rapamycin disposition in human tissue homogenates and Caco-2 cell monolayers. Metabolism of Seco Rapamycin (D2) to dihydro Sirolimus (M2) is tested by incubating 20μM Seco Rapamycin with human liver, jejunal mucosal, and Caco-2 homogenates. All the homogenates generate M2 in an NADPH-dependent manner. Ketoconazole at a high concentration (100μM) has no effect on M2 formation in any of the homogenates examined. Metabolism of Seco Rapamycin to M2 in intact cells is tested by adding 20μM Seco Rapamycin to Caco-2 cell monolayers. With apical application, only a little Seco Rapamycin is found in the basolateral compartment and cellular fraction after 4 h, and little M2 is detected either. LY335979 has little effect on Seco Rapamycin distribution after an apical dose, although M2 becomes detectable in the apical compartment. By contrast, with basolateral application of Seco Rapamycin, Seco Rapamycin and M2 are both readily found in the apical compartment; LY335679 lowers Seco Rapamycin flux toward the apical compartment while raising M2 amounts in the apical and basolateral compartments[1].
