Background

SIGLEC10 (sialic acid-binding immunoglobulin-like lectin 10) is an inhibitory receptor of the Siglec family expressed mainly on immune cells including macrophages, dendritic cells, and B cells. Through ITIM motifs in its cytoplasmic tail, SIGLEC10 transduces inhibitory signals upon engaging sialylated glycan ligands. It binds CD24 and the alarmin HMGB1, contributing to recognition of self-associated molecular patterns and dampening of inflammatory responses. By interacting with sialoglycans overexpressed on tumor cells, the CD24-SIGLEC10 axis promotes a 'don't eat me' signal that suppresses macrophage phagocytosis, making SIGLEC10 an emerging target in cancer immunology and innate immune regulation.

The SIGLEC10 ORF cDNA Lentivirus delivers the full human SIGLEC10 open reading frame for stable overexpression in mammalian cells. The ORF is expressed from a constitutive CMV promoter within a third-generation, VSV-G-pseudotyped lentiviral vector. Depending on configuration, constructs may include a C-terminal epitope tag and a fluorescent reporter (EGFP, GFP, mCherry, or RFP) and/or an antibiotic selection marker (puromycin or blasticidin), with tag, reporter, and selection elements fused or linked by self-cleaving peptide sequences for independent protein expression. The system is used for gain-of-function studies of SIGLEC10 in immune signaling, macrophage regulation, and tumor immune evasion, and to generate stable cell lines by antibiotic selection or FACS sorting. Supplied as ultra-purified, high-titer lentiviral particles suitable for primary and difficult-to-transfect cells.

About This Product

This ORF cDNA lentivirus enables stable overexpression of SIGLEC10 (NCBI Accession: NM_033130) in mammalian cells via a third-generation, VSV-G pseudotyped delivery system. The ORF cDNA is fused to a C-terminal epitope tag (V5, Myc, or HA) and expressed under a strong constitutive promoter (CMV). Reporter and selection marker components (EGFP, GFP, mCherry, RFP; Blasticidin, Puromycin) are co-expressed via self-cleaving P2A peptides, enabling independent protein production without fusion-tag artifacts.

Ultra-purification by PEG precipitation and sucrose gradient centrifugation yields high-titer particles suitable for primary cells, suspension cultures, and stem cells. Stable polyclonal cell lines are established within 10–14 days by antibiotic selection or FACS sorting. For in vivo applications, the serum-free formulation and VSV-G envelope support direct administration or further concentration for stereotactic injection.
