Target Sin Nombre Virus

Scientific Background

Sin Nombre Virus (SNV) is a New World hantavirus and the principal etiologic agent of Hantavirus Pulmonary Syndrome (HPS) in North America, first identified during a 1993 outbreak in the U.S. Southwest. SNV is transmitted to humans through aerosolized excreta of infected deer mice (Peromyscus maniculatus). The Gn glycoprotein is the major surface envelope protein of hantaviruses, forming heterodimeric complexes with Gc on the virion surface. Gn mediates receptor binding via β3 integrins, drives viral entry, and is the primary target of neutralizing antibodies. Its conservation across hantavirus strains makes SNV Gn a critical antigen for serodiagnosis, epidemiological surveillance, and hantavirus vaccine research.

Product Description

Hantavirus

This recombinant Sin Nombre Virus Glycoprotein (Gn) minus the transmembrane domain (SNVGnΔTM) is a HIS-tagged protein produced in mammalian cells and purified by FPLC. Supplied as purified liquid protein in formulation buffer.

Functional Activity

SNVGnΔTM has been validated by capture ELISA and Western Blot. In a capture ELISA, SNVGnΔTM was coated at 2 µg/mL in PBS and detected with a mouse anti-Sin Nombre Virus GP polyclonal antibody (IBT Cat# 0309-001), demonstrating a specific dose-response. SDS-PAGE analysis confirms protein integrity under denaturing and reducing conditions; Western Blot detection was performed using anti-HIS antibody at 1:1000 dilution.

Safety & Handling

Research Use Only (RUO). Not approved for human or veterinary use. Handle according to institutional biosafety guidelines. Commercial use requires written approval from Integrated BioTherapeutics, LLC.

What expression system was used to produce this protein?

SNVGnΔTM is produced in mammalian cells and purified by FPLC. Mammalian expression preserves the eukaryotic post-translational modifications — including glycosylation — important for native viral glycoprotein antigenicity.

What applications has this protein been validated for?

SNVGnΔTM has been validated for capture ELISA and Western Blot by IBT Bioservices. In ELISA, the protein was coated at 2 µg/mL in PBS and detected with a mouse anti-SNV GP polyclonal antibody. In Western Blot, the HIS-tag was detected with anti-HIS antibody at 1:1000 dilution under denaturing and reducing conditions.

What is the recommended ELISA coating concentration?

Per the IBT Bioservices datasheet, SNVGnΔTM can be coated at 2 µg/mL in PBS for capture ELISA. Optimal concentrations should be determined empirically for each assay system.

How should this protein be stored?

Store at -20°C for short-term use (up to 1 week). For long-term storage, keep at -80°C. Upon receipt, aliquot into single-use volumes to avoid repeated freeze-thaw cycles. The formulation buffer (20 mM TrisHCl pH 8, 0.15 M NaCl, 5% Sucrose) provides cryoprotection at -80°C.
