Product Overview
The Spin-Column Protein Folding Screen Kit is designed to produce active proteins from guanidine hydrochloride- or urea-solubilized inclusion bodies, or from protein aggregates formed during expression, purification or storage. Different proteins fold under different conditions, so the kit provides 10 spin columns representing 10 diverse folding conditions, including conditions that allow disulfide bond formation and reducing conditions.
The folded samples are checked by SDS-PAGE and activity tests, and the best condition (column number) is then used for preparative folding with the matching Preparative Protein Folding Column Set.

Protocol Outline
- Solubilize inclusion bodies in 20 mM Tris-HCl, pH 8.0, 6 M guanidine hydrochloride (or 8 M urea), 10 mM DTT, and clear by centrifugation.
- Keep the columns and reagents at 2–8°C. Mix 130 µl of solubilized inclusion bodies with 130 µl of Solution A and incubate for 5 min.
- Spin the columns at 3200 rpm for 1 min, remove the bottom tips and caps, spin at 1400 rpm for 2 min and move each column to a clean tube.
- Load 25 µl of the loading sample onto each column and spin at 3200 rpm for 4 min; incubate the eluates at 4°C for 2–4 h, add 50 µl Solution B, incubate at 4°C for 2 h to overnight, spin at 14,000 rpm for 5 min and analyze the supernatant by SDS-PAGE and activity tests.
The ProFoldin protocol “Spin-Column Protein Folding Screen” is linked under Documents.
Notes
- Catalog # SFC01-10 is this screen kit: 10 spin columns, each a different folding condition (#1–10). Catalog #s SFC01 to SFC10 (without -10) are the small-scale preparative sets of the Preparative Protein Folding Column Set, each with 10 identical columns of one selected condition.
