Product Overview

This kit is suitable for separating total DNA from 180–220 mg of fresh or frozen human or animal feces. The genomic DNA, viral DNA, bacteria and parasite DNA of the human or animal in the dissolved feces can be bound to the nucleic acid purification column, and degraded protein and PCR inhibitors are removed by the Solution InR precipitation and the column wash steps, and the genomic DNA is washed with Solution RP and Wash Buffer. After washing, it can be used in various molecular biology experiments by eluting with the supplied Elution Buffer.

The sample is lysed in 1.4 mL of Solution SGE in a 95°C water bath for 5 min, clarified with one fifth volume of Solution InR, then digested with 10 µL of Proteinase K and 200 µL of Solution GA2 in a 70°C water bath for 10 min. After adding 200 µL of absolute ethanol the mixture is loaded onto Adsorption column G, washed with 500 µL of Solution RP and 700 µL of Wash Buffer, dried by centrifugation at 12,000 rpm for 2 min, and eluted with 50–200 µL of Elution Buffer after 2–5 min at room temperature.

Usage Notes

  • Store freshly collected fecal samples at -20°C or lower without delay; 2–3 hours at room temperature causes degradation and longer storage risks severe DNA degradation.
  • Repeating the Wash Buffer step increases purity.
  • Remove residual ethanol by centrifuging at 12,000 rpm for 2 min and air-drying the column, to avoid inhibition of downstream enzymes.
  • For sensitive assays, elute with sterile water at pH 7.0–8.5 instead of Elution Buffer.
  • A second elution with a further 50–200 µL increases yield.
  • Store the eluted DNA at -20°C.
