In Vitro
The pKa values of TES (0.1 M) sodium are 7.92 at 0°, 7.50 at 20°, and 7.14 at 37°, and a saturated aqueous solution of it reaches 2.6 M at 0°[1]. In a closed culture environment, TES (6-50 mM; 11-14 days) sodium is similar to, or even more effective than, NaHCO3-CO2-buffered medium regarding the efficiency of colony formation, the size of colonies, and cell morphology across several mammalian cell lines (HeLa S3, KB, BHK 21-Cl3, 3Y1-B Cl-1)[2]. In the SV40 virus plaque assay with monkey kidney CV-1 cells, TES (25-50 mM; 14-19 days) sodium shows efficiency similar to NaHCO3-CO2-buffered medium, with a shorter required time[2].
