Thapsigargin is a potent inhibitor of the sarco/endoplasmic reticulum Ca²⁺-ATPase (SERCA), leading to depletion of ER calcium stores and induction of ER stress. In neuroscience, Thapsigargin is used to model ER stress-related neuronal dysfunction, a key feature in neurodegenerative diseases such as Alzheimer’s and Parkinson’s. ER stress triggers the unfolded protein response (UPR), which can lead to apoptosis if unresolved. Thapsigargin is instrumental in studying the molecular pathways of calcium homeostasis, protein misfolding, and cell death in neurons.
