Thiol Fluorometric Assay Kit

SKU:BHT19600014
Overview
Click light‑blue chips for details
Free thiols, overwhelmingly contributed by glutathione and protein cysteines, buffer the cell against oxidative insult. This kit quantifies Thiol by fluorescence at Ex/Em = 400/465 nm. Suitable for cells, tissues. Measuring total free thiol captures both glutathione and protein cysteines, giving a broader picture of redox buffering than GSH alone.

About the Analyte

Free thiols, overwhelmingly contributed by glutathione and protein cysteines, buffer the cell against oxidative insult, and their depletion is one of the earliest measurable signs of redox imbalance.

Assay Principle

The Thiol Fluorometric Assay Kit applies a fluorometric format to this measurement, read at Ex/Em = 400/465 nm. Hands-on and incubation time is 10 minutes.

Thiols are extremely efficient antioxidants that can protect cellular lipids, proteins, and nucleic acids against peroxidative damage due to their strong reductive capacity and their ability to react with free radicals.

Measuring total free thiol captures both glutathione and protein cysteines, giving a broader picture of redox buffering than GSH alone.

Q.What instrument do I need to quantify this analyte?
A.This kit requires a fluorescence-capable plate reader. The readout is taken at Ex/Em = 400/465 nm.
Q.What does the measured level tell me?
A.Measuring total free thiol captures both glutathione and protein cysteines, giving a broader picture of redox buffering than GSH alone.
Q.Why measure Thiol?
A.Free thiols, overwhelmingly contributed by glutathione and protein cysteines, buffer the cell against oxidative insult, and their depletion is one of the earliest measurable signs of redox imbalance.
Q.Which sample matrices are validated?
A.The datasheet lists cells, tissues. Other matrices can usually be adapted, but run a spike-recovery check before relying on the values.
Q.How long does the quantitation take?
A.Combined incubation and handling time is 10 minutes. Keep this interval identical across plates in the same study, since timing differences show up directly in the endpoint.
Q.What is the detection range?
A.The kit is validated for 0.3-20 µM under datasheet conditions. Dilute samples that read above the range and concentrate those below it rather than extrapolating from the standard curve.
