In Vitro
TLR7/8 agonist 1 displays prominent immunostimulatory activities and is a convenient precursor for covalent attachment of fluorophores, with no significant loss of activity. TLR7/8 agonist 1 retains TLR7-agonistic activity, with an EC50 of 20 nM, and is coupled directly and covalently to commercially available fluorescein isothiocyanate and rhodamine B isothiocyanate[1]. In primary screens, TLR7/8 agonist 1 shows substantially different agonistic potencies for human TLR7 (50 nM) and TLR8 (55 nM)[2].

Cell Assay[2]
Isolate fresh human peripheral blood mononuclear cells (hPBMC) on Ficoll–Hypaque gradients from human blood collected by venipuncture, with informed consent and following institutional guidelines. Stimulate aliquots of PBMCs (105 cells in 100 μL/well) for 12 h with test compounds at graded concentrations (e.g., TLR7/8 agonist 1; 0.1, 1, 10, and 100 μg/mL). Isolate the supernatants by centrifugation and assay them in duplicates with analyte-specific multiplexed cytokine/chemokine bead array assays[2].
