In Vitro
Mutations that help tumors evade immune surveillance invariably accompany tumor growth. Even so, tumorigenesis causes mutated antigens, specifically neoantigens, to accumulate, and after ex vivo stimulation the host immune system readily recognizes them. Delivering these neoantigens to activated dendritic cells through antibody-tumor immune complexes therefore offers a way to overcome the state of "immune ignorance" of the immune system toward the tumor. Indeed, intratumoral injection that delivers tumor-binding antibodies together with dendritic cell adjuvants has successfully induced potent anti-tumor immune responses. Dendritic cell adjuvants, such as Toll-like receptor agonists, thus represent a key potential strategy against cancer[1].
