Monocyte Chemotactic Protein-1 (MCP-1), also known as Chemokine (C-C motif) ligand 2 (CCL2), is a member of the beta (CC) subfamily of chemokines. MCP-1 is characterized as a monocyte-specific chemoattractant. Because of its target cell specificity, MCP-1 is postulated to play a pathogenic role in a variety of diseases characterized by mononuclear cell infiltration, including atherosclerosis, rheumatoid arthritis and allergic responses. In basophils, MCP-1 is highly effective as a stimulus of histamine release but has only weak chemotactic activity. Additionally, it has been shown to chemoattract CD4+ and CD8+ T lymphocytes.
Assay Principle
The Mouse MCP-1 ELISA is an enzyme-linked immunosorbent assay for the quantitative detection of mouse MCP-1 in different tissue plasma, serum, and other biological samples. The plate has been coated with murine MCP-1 monoclonal antibody. Any MCP-1 present in the sample or standard will be captured. A biotin-conjugated anti-murine MCP-1 antibody is added followed by streptavidin-horseradish peroxidase (HRP) and an accompanying substrate. The reaction exhibits a blue color in direct proportion to the amount of MCP-1 in the initial sample. The reaction is terminated by the addition of a stop solution, and absorbance is measured at 450 nm. The concentration of MCP-1 in the samples is calculated opposite to the standard curve.
Tribo™ Mouse CCL2/MCP-1 ELISA Kit is a Solid-phase sandwich ELISA for the quantitative determination of CCL2/MCP-1. The microplate is pre-coated with a capture antibody specific to the target. Target present in the sample or standard binds the immobilized antibody, is detected with a biotinylated detection antibody and streptavidin-HRP, and quantified colorimetrically against a standard curve.
Kit Components
The kit provides a pre-coated microplate, target standard for standard-curve generation, detection reagents (detection antibody and enzyme conjugate), wash buffer, substrate, and stop solution. Refer to the product datasheet for the complete component list and quantities.

Safety & Handling
This kit is validated for serum, plasma, tissue. Other matrices may require additional in-lab validation.
This assay has a sensitivity of < 8.0 pg/mL. Samples reading outside the range should be diluted or concentrated.
The kit provides a pre-coated microplate, target standard for standard-curve generation, detection reagents (detection antibody and enzyme conjugate), wash buffer, substrate, and stop solution. See the datasheet for the full component list and quantities.
A standard microplate reader capable of measuring absorbance at 450 nm is required for this sandwich ELISA.
