Compound Overview
Trimethyl chitosan, also known as N,N,N-Trimethylchitosan, is a multifunctional polymer derived from Chitosan. It targets the absorption-enhancing proteins of tight junctions in intestinal and mucosal epithelial cells, inducing tight junction protein rearrangement and increasing intercellular permeability. It can stimulate transmembrane transport of hydrophilic drugs such as peptides and proteins, and it can be used for drug delivery and the synthesis of nanoparticles[1][2]. It is supplied as an off-white to light yellow solid.

Safety
For Research Use Only. Not for use in diagnostic or therapeutic procedures, and not for human or veterinary use. Handle in accordance with the Safety Data Sheet and your institution's chemical hygiene plan.
In Vitro
| Solvent | Solubility | Notes |
|---|---|---|
| H2O | 33.33 mg/mL | requires sonication and warming and heat to 60°C |
Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); avoid repeated freeze-thaw cycles.
In Vivo
Choose the formulation that suits the animal model and route of administration. Percentages are volume ratios of the final working solution. Prepare the working solution fresh on the day of dosing; if precipitation or phase separation occurs, gentle warming or sonication can help.
Direct preparation of the working solution
These formulations are prepared directly, without a DMSO stock; use them promptly after preparation.
Protocol 1
| Composition | PBS |
|---|---|
| Result | 10 mg/mL; clear solution; requires sonication |

In Vitro
In Caco-2 cell monolayer experiments, Trimethyl chitosan lowered transepithelial electrical resistance (TEER) concentration-dependently at 0.05-1.0% (w/v) over 2-4 h, and significantly raised the transmembrane transport efficiency of 14C-mannitol, while cell viability was not significantly affected[1].
