The Tte UvrD Helicase, derived from the thermophilic bacterium Thermoanaerobacter tengcongensis, possesses the ability to unwind double-stranded DNA under high-temperature conditions. This enzyme is stable at 65°C and can be applied in isothermal amplification techniques without relying on single-strand binding proteins. In thermophilic Helicase-Dependent Amplification (tHDA), it assists polymerase in DNA strand separation, enabling isothermal amplification.

In the LAMP system, it effectively eliminates non-specific amplification. Two sets of LAMP primers were used to prepare LAMP reaction systems, with each set including an experimental group supplemented with 10 ng Tte UvrD Helicase and a control group without it.

Free of exonuclease, endonuclease, and RNase contamination. 20 ng of Tte UvrD Helicase was incubated with nucleic acid substrates, and band changes were analyzed via agarose gel electrophoresis. Experimental results showed that no exonuclease, endonuclease, or RNase contamination was detected in three batches of Tte UvrD Helicase.

Store this enzyme at -20°C and avoid repeated freeze-thaw cycles to preserve catalytic activity. The product is shipped Dry Ice and remains stable for up to one year from the date of manufacture when stored under recommended conditions. Aliquoting the stock solution into single-use volumes is recommended for enzymes used infrequently to minimize thermal cycling of the bulk stock.
This enzyme is validated for use in Isothermal Amplification applications. It is suitable for use in both standard laboratory research settings and, where applicable, optimized reaction workflows requiring high specificity, sensitivity, or throughput.
Helicase activity (µg/µL) is validated by strand displacement activity on partially double-stranded DNA substrates under isothermal conditions at 37°C.
This enzyme is produced as Recombinant (E. coli) and supplied as a Research Use Only (RUO) reagent. Each lot is subjected to activity assay, purity assessment by SDS-PAGE, and functional validation prior to release. A Certificate of Analysis (CoA) and Safety Data Sheet (SDS) are available on request.
Helicase-dependent amplification uses a thermostable UvrD-family or ATP-dependent helicase to unwind double-stranded DNA at isothermal temperatures (60–65°C), creating single-stranded templates for primer annealing and extension. Unlike PCR, HDA requires no thermal cycling equipment, making it suitable for portable or point-of-care diagnostics. SSB protein stabilizes separated strands to prevent re-annealing during amplification.
