Product Overview
U-Blot® Prestained Protein Ladder (10-180 kDa) contains 10 highly purified prestained recombinant proteins (10, 15, 25, 35, 40, 55, 70, 100, 130, 180 kDa). Reference bands are color-coded (70 kDa orange-red, 10 kDa green).
The broad molecular-weight range allows dynamic observation of protein migration during electrophoresis and precise estimation of target-protein size. After SDS-PAGE, the prestained bands transfer to PVDF or NC membranes as clearly visible colored bands, allowing transfer efficiency to be monitored directly during Western blotting.

Applications
Designed for SDS-PAGE polyacrylamide gel electrophoresis and Western blotting, serving as a molecular-weight reference and a visual indicator of transfer performance.
Compatibility & Usage Notes
Ready to use: do not boil, dilute, or add reducing agents. Prestained proteins show different apparent molecular weights in different buffer systems; use as a reference relative to unstained standards when precise sizing is required.
Safety & Handling
Instructions for Use
- This product is ready to use. Before adding the sample, place it at room temperature for a few minutes to thaw, then gently shake to ensure the solution is evenly mixed. Do not boil, dilute, or add reducing agents.
- Load 5 µl of the sample into the gel well for polyacrylamide gel electrophoresis (increase the volume for wide wells).
Technical Notes
- Large proteins (~100 kDa) may need a longer transfer time or higher current. Avoid adding SDS to the transfer buffer; if required, keep it below 0.05%. For best transfer, consider U-Blot® Fast Transfer Buffer (Cat. No. W2206).
- Prestained bands migrate at different apparent rates in various SDS-PAGE buffers; in low-percentage gels (<10%) low-MW bands may migrate with the dye and should be used only as a reference.
- A 12% separating gel or 4-20% gradient gel is recommended.
Migration Pattern
Migration pattern of U-Blot® Prestained Protein Molecular Weight Markers. The apparent molecular weight (kDa) of each protein is determined by calibration against unstained protein standards in various electrophoresis buffer systems.
Yes. Thaw at room temperature for a few minutes and mix gently before loading. Do not boil, dilute, or add reducing agents.
It spans 10-180 kDa across 10 bands (10, 15, 25, 35, 40, 55, 70, 100, 130, 180 kDa).
70 kDa orange-red, 10 kDa green, providing visual reference points on the gel and membrane.
Approximately 5 µl per lane; increase slightly for wide wells. Run a pilot to optimize for your system.
Stable at -20°C for 36 months; 4°C for 3 months; 25°C for 4 weeks Aliquot and avoid repeated freeze-thaw for best performance.
