Product Overview

U-Blot® Prestained Protein Ladder (10-250 kDa) contains 9 highly purified prestained recombinant proteins (10, 15, 25, 35, 55, 70, 100, 130, 250 kDa). Reference bands are color-coded (25 kDa and 70 kDa orange-red, 10 kDa green).

The broad molecular-weight range allows dynamic observation of protein migration during electrophoresis and precise estimation of target-protein size. After SDS-PAGE, the prestained bands transfer to PVDF or NC membranes as clearly visible colored bands, allowing transfer efficiency to be monitored directly during Western blotting.

Applications

Designed for SDS-PAGE polyacrylamide gel electrophoresis and Western blotting, serving as a molecular-weight reference and a visual indicator of transfer performance.

Compatibility & Usage Notes

Ready to use: do not boil, dilute, or add reducing agents. Prestained proteins show different apparent molecular weights in different buffer systems; use as a reference relative to unstained standards when precise sizing is required.

Safety & Handling

For Research Use Only. Not for use in diagnostic procedures. Store as indicated; aliquot to avoid repeated freeze-thaw cycles.

Instructions for Use

  • This product is ready to use. Before adding the sample, let it stand at room temperature for several minutes to thaw, then gently mix. Do not boil, dilute, or add a reducing agent.
  • Load 5 µl into the gel well for polyacrylamide gel electrophoresis (increase for wide wells). For first-time use, run a pilot to determine the optimal load.
  • For frequent use it may be stored at 2-8°C (3-month validity); aliquot to avoid repeated freeze-thaw.

Technical Notes

  • Large proteins (~100 kDa) may need a longer transfer time or current. Avoid SDS in the transfer buffer; keep below 0.05% if required.
  • Prestained bands migrate differently in various SDS-PAGE buffers; in low-percentage gels (<10%) low-MW bands may migrate with the dye and are for reference only.
  • A 12% separating gel or 4-20% gradient gel is recommended.
  • Wear a lab coat and disposable gloves when handling.

Migration Pattern

Migration pattern of U-Blot® Prestained Protein Ladder (10-250 kDa): apparent molecular weights (kDa) are determined by calibration against unstained protein standards in various electrophoresis buffer systems.

Is this ladder ready to use?

Yes. Thaw at room temperature for a few minutes and mix gently before loading. Do not boil, dilute, or add reducing agents.

What molecular-weight range and how many bands does it cover?

It spans 10-250 kDa across 9 bands (10, 15, 25, 35, 55, 70, 100, 130, 250 kDa).

Which bands are colored for orientation?

25 kDa and 70 kDa orange-red, 10 kDa green, providing visual reference points on the gel and membrane.

How much should I load per lane?

Approximately 5 µl per lane; increase slightly for wide wells. Run a pilot to optimize for your system.

How should I store it and what is the shelf life?

Stable at -20°C for 36 months; 4°C for 3 months; 25°C for 4 weeks Aliquot and avoid repeated freeze-thaw for best performance.
