UDG (uracil DNA glycosylase) can catalyze the hydrolysis of the N-glycosidic link between the uracil base and the sugar-phosphate backbone in ssDNA and dsDNA, it has no catalytic activity in RNA.
This product has been processed by the UCF.ME™ ultra-low residue process, and its residual DNA contamination of E. coli is extremely low, which is suitable for applications with stricter requirements for background bacteria and higher performance demands, such as the development of real-time quantitative PCR systems that support the pre-mixing of primers and probes.

Store this enzyme at -20°C and avoid repeated freeze-thaw cycles to preserve catalytic activity. The product is shipped Dry Ice and remains stable for up to one year from the date of manufacture when stored under recommended conditions. Aliquoting the stock solution into single-use volumes is recommended for enzymes used infrequently to minimize thermal cycling of the bulk stock.
This enzyme is validated for use in PCR & Amplification, Molecular Diagnostics applications. It is suitable for use in both standard laboratory research settings and, where applicable, optimized reaction workflows requiring high specificity, sensitivity, or throughput.
One unit (U) is defined as the amount of enzyme that catalyzes the release of 1 nmol of uracil from a defined uracil-containing single-stranded DNA substrate in 30 min at 37°C.
This enzyme is produced as Recombinant (E. coli) and supplied as a Research Use Only (RUO) reagent. Each lot is subjected to activity assay, purity assessment by SDS-PAGE, and functional validation prior to release. A Certificate of Analysis (CoA) and Safety Data Sheet (SDS) are available on request.
Uracil-DNA Glycosylase (UDG/UNG) cleaves the N-glycosidic bond between uracil bases and the sugar-phosphate backbone of DNA containing deoxyuridine (dU). In carryover prevention protocols, dUTP is substituted for dTTP in PCR master mixes. Any carryover amplicon from previous reactions contains dU and is degraded by UDG treatment before PCR cycling, preventing false-positive results. The UDG is inactivated during the initial denaturation step, allowing normal amplification of native dT-containing template DNA.
