UCF.ME Uracil DNA Glycosylase derived from psychrophilic marine bacteria, it is an ultra-low residual heat-labile UDG enzyme known as UCF.METM. This product can prevent the residual activity of conventional UDG enzymes after inactivation from degrading dU-containing amplification products at room temperature. It also prevents background bacteria present in molecular enzymes from causing false positives in PCR results. Therefore, the introduction of UCF.METM ultra-low residual thermolabile UDG enzyme is crucial for accurately identifying infectious pathogens and assisting in the clinical diagnosis of infections and infectious diseases.

Features

Ultra-low residual of UCF.METME. coli genomic DNA residue <0.1 copies/ U;

Low nuclease residueNo residual exonuclease, nicking enzyme, or RNase;

Strong digestion capability0.05 U/T can digest 105 copies/T dU-DNA products;

Good thermolabilityComplete inactivation under any condition of 50for 10 minutes, 55for 5 minutes, or 95for 5 minutes;

Compatible with RT-qPCR reaction systemsNo impact on the detection system even at high input levels (2U/20μL reaction system).

Figures

Figure 1. 14466ES protein purity (SDS-PAGE) detection results. Protein Purity≥95%

Figure 2. 14466ES Nuclease residue test results. No Residual Nucleases, No Residual Exonucleases, Nicking Enzymes, or RNases.

Figure 3. 14466ES E.coli genomic DNA residue test results. E.coli genomic DNA residue< 0.1 copies/ U, the residue levels were significantly lower than the conventional UDG.

Figure 4. Strong digestive capability. 0.05 U/T is sufficient to digest 10^5 copies/T of dU-DNA products. 14466ES was used to prepare qPCR mix, while the control group of qPCR mix without UDG enzyme could completely digest 105 copies/T dU-DNA products.

Figure 5. After 50℃, 10min; 55℃, 5min; 55℃, 10min; 95℃, 5min heat inactivation treatment, 14466ES lost digestive ability.

Store this enzyme at -20°C and avoid repeated freeze-thaw cycles to preserve catalytic activity. The product is shipped Dry Ice and remains stable for up to one year from the date of manufacture when stored under recommended conditions. Aliquoting the stock solution into single-use volumes is recommended for enzymes used infrequently to minimize thermal cycling of the bulk stock.

This enzyme is validated for use in PCR & Amplification, Molecular Diagnostics applications. It is suitable for use in both standard laboratory research settings and, where applicable, optimized reaction workflows requiring high specificity, sensitivity, or throughput.

One unit (U) is defined as the amount of enzyme that catalyzes the release of 1 nmol of uracil from a defined uracil-containing single-stranded DNA substrate in 30 min at 37°C.

This enzyme is produced as Recombinant (E. coli) and supplied as a Research Use Only (RUO) reagent. Each lot is subjected to activity assay, purity assessment by SDS-PAGE, and functional validation prior to release. A Certificate of Analysis (CoA) and Safety Data Sheet (SDS) are available on request.

Uracil-DNA Glycosylase (UDG/UNG) cleaves the N-glycosidic bond between uracil bases and the sugar-phosphate backbone of DNA containing deoxyuridine (dU). In carryover prevention protocols, dUTP is substituted for dTTP in PCR master mixes. Any carryover amplicon from previous reactions contains dU and is degraded by UDG treatment before PCR cycling, preventing false-positive results. The UDG is inactivated during the initial denaturation step, allowing normal amplification of native dT-containing template DNA.
