Compound Overview
WH-15 is a fluorogenic PLC reporter with Km values of 49, 30, and 86.1 μM for PLC-γ1, PLC-δ1, and PLC-β2, respectively. It can be cleaved in a cascade reaction to generate fluorescent 6-aminoquinoline, and it can be used to respond to imaging PLC activity in live cells (Ex/Em = ~344/530 nm)[1]. It is supplied as a light yellow to yellow solid (C31H43N2O18P3, MW 824.60) at 99.51% purity.

In Vitro
Guide (this recommended protocol is a guideline only and can be adjusted to specific needs)[1]. 1. Solution preparation 1.1 Stock solution preparation Solvent: DMSO. Concentration: 25 mM (optimize per experiment). 1.2 Working solution preparation Dilute to 50 μM in assay buffer. Note: Prepare the working solution and use it immediately, protected from light. 2. Cell staining 2.1 Transfect HEK-293T cells with the indicated DNA (600 ng) and empty vector (100 ng). 2.2 At 37°C, add 70 μL of 50 μM WH-15 (in 2 mM DTT, 3 mM EGTA, 3 mM CaCl2, 70 mM KCl, 50 mM HEPES (pH 7.2) and 83 μg/mL fatty-acid free BSA) to 50 μL of normalized cell lysate. 2.3 Collect data every 3 min using a Multilabel Counter (Ex/Em = 340/535 nm).
