Compound Overview

XX-650-23 is a potent CREB inhibitor that blocks CREB function by disrupting the CBP-CREB interaction, and it can be used in AML research[1]. It is supplied as an off-white to yellow solid (C18H12N2O2, MW 288.30) at 99.90% purity.

Safety

For Research Use Only. Not for use in diagnostic or therapeutic procedures, and not for human or veterinary use. Handle in accordance with the Safety Data Sheet and your institution's chemical hygiene plan.

In Vitro

SolventSolubilityNotes
DMSO100 mg/mL (346.86 mM)requires sonication; use freshly opened DMSO (absorbed moisture lowers solubility)

Aliquot the stock solution and store it at -80°C (up to 6 months) or -20°C (up to 1 month); avoid repeated freeze-thaw cycles.

In Vivo

Choose the formulation that suits the animal model and route of administration; percentages are volume ratios of the final working solution. Start from a clear DMSO stock (see In Vitro above), add the co-solvents one at a time in the order listed, mixing after each addition, and prepare the working solution fresh on the day of dosing. If precipitation or phase separation occurs, gentle warming or sonication can help.

Protocol 1

Composition10% DMSO + 90% Corn Oil
Result2.5 mg/mL (8.67 mM); clear solution; requires sonication
How to prepareGives a clear solution at 2.5 mg/mL. Use with caution if continuous dosing will exceed two weeks. For 1 mL of working solution: add 100 μL DMSO stock (25.0 mg/mL) to 900 μL corn oil.

In Vitro

AML cell growth is inhibited by XX-650-23 (870 nM-2.3 μM; 48 hours), with IC50 values of 870 nM (HL-60), 910 nM (KG-1), 2.0 μM (MOLM-13) and 2.3 μM (MV-4-11)[1]. XX-650-23 (5 μM; 6 or 24 hours) lowers H3K27 acetylation specifically, without acting as a general inhibitor of acetyltransferase activity[1]. XX-650-23 (2 μM; 72 hours) triggers apoptosis via the intrinsic apoptosis pathway, with caspase-3 activation and detectable caspase-9 cleavage, and also downregulates Mcl-1 and bcl-2[1].
