YE6144 is a selective, prototypical inhibitor of interferon regulatory factor 5 (IRF5). It suppresses disease course and is especially effective for remission maintenance in a mouse model of systemic lupus erythematosus (SLE), and can be used in SLE research[1]. Start from a clear DMSO stock (see In Vitro above), add the co-solvents one at a time in the order listed, mixing after each addition, and prepare the working solution fresh on the day of dosing. If precipitation or phase separation occurs, gentle warming or sonication can help.

How to prepareGives a clear solution at ≥ 10 mg/mL (saturation not determined). For 1 mL of working solution: add 100 μL DMSO stock (100.0 mg/mL) to 400 μL PEG300; then 50 μL Tween-80; then 450 μL saline to bring the volume to 1 mL. Saline: dissolve 0.9 g sodium chloride in ddH2O and make up to 100 mL.

In Vitro

YE6144 (1 or 3 μM; 30 min) inhibits IRF5 phosphorylation in both human PBMCs and mouse splenocytes[1]. In human HC PBMCs, YE6144 (0-10 μM; 30 min) inhibits type I IFN production, with an IC50 of approximately 0.09 μM[1].
