Overview

Multifunctional transcription factor that exhibits positive and negative control on a large number of cellular and viral genes by binding to sites overlapping the transcription start site. Binds to the consensus sequence 5'-CCGCCATNTT-3'; some genes have been shown to contain a longer binding motif allowing enhanced binding; the initial CG dinucleotide can be methylated greatly reducing the binding affinity. The effect on transcription regulation is depending upon the context in which it binds and diverse mechanisms of action include direct activation or repression, indirect activation or repression via cofactor recruitment, or activation or repression by disruption of binding sites or conformational DNA changes. Its activity is regulated by transcription factors and cytoplasmic proteins that have been shown to abrogate or completely inhibit YY1-mediated activation or repression. For example, it acts as a repressor in absence of adenovirus E1A protein but as an activator in its presence. Acts synergistically with the SMAD1 and SMAD4 in bone morphogenetic protein (BMP)-mediated cardiac-specific gene expression. Binds to SMAD binding elements (SBEs) (5'-GTCT/AGAC-3') within BMP response element (BMPRE) of cardiac activating regions. May play an important role in development and differentiation. Proposed to recruit the PRC2/EED-EZH2 complex to target genes that are transcriptional repressed. Involved in DNA repair. In vitro, binds to DNA recombination intermediate structures (Holliday junctions). Proposed core component of the chromatin remodeling INO80 complex which is involved in transcriptional regulation, DNA replication and probably DNA repair; proposed to target the INO80 complex to YY1-responsive elements. [UniProt]

This anti-YY1 antibody is supplied as Purified (Rabbit, Polyclonal (rabbit origin), Rabbit Ig, Unconjugated) and is designed to support common target-detection workflows after the on-page specifications.

Key elements and design rationale

  • Target: YY1
  • Format: Purified
  • Species reactivity: Human
  • Applications (listed): WB
  • Conjugate: Unconjugated
  • Clone and antibody class: Polyclonal (rabbit origin), Rabbit Ig

Because antibody performance can depend on epitope context, sample preparation, and biological state, interpret signals using appropriate controls and orthogonal evidence when possible.

Biological background

YY1 is referenced in public gene/protein resources (e.g., UniProt and NCBI Gene), which provide curated names/synonyms, protein features, and pathway context. When designing assays, consider potential isoforms, post-translational modifications, and cell-type specific expression that may influence observed signal.

Research relevance and current trends

  • Profiling YY1 expression across model systems, perturbations, and time points to support mechanistic hypotheses.
  • Combining antibody-based detection with multi-omics or imaging readouts to link YY1 signal with phenotype.
  • Using well-matched controls (isotype controls, genetic perturbations, or independent reagents) to strengthen interpretation of target-associated signal.

Common research applications

  • WB

Use the listed applications as a starting point and tailor experimental design to your sample type and readout requirements.

Notes for experimental interpretation

  • Specificity considerations: closely related family members, isoforms, or PTMs can affect apparent specificity; confirm with independent approaches when critical.
  • Controls: include negative controls and, when feasible, genetic or pharmacologic perturbations to support target attribution in your system.
  • Species and sample context: differences in sequence, expression, fixation, or extraction conditions can change signal behavior across models.
