U2OS began with tibial tumor tissue taken from a 15-year-old female patient; the human osteosarcoma line that resulted now ranks among the most heavily used cell models in cancer biology and molecular genetics. The cells attach and adopt an epithelial-like shape, and they turn up throughout work on cell cycle regulation, DNA damage response, genome stability, signal transduction, gene expression, and cancer therapeutics. p53 function in the line remains wild-type, which is what makes it so useful for digging into tumor suppression mechanisms and cellular stress responses. GenJet™ DNA In Vitro Transfection Reagent for U2OS Cells arrives with its conditions already tuned, so DNA transfection of U2OS cells runs efficiently; the protocol below was refined to reach maximal efficiency in these cells while viability stays excellent.
