Product Overview

Built as an enhanced formulation of the original GenJet™ In Vitro DNA Transfection Reagent, GenJet™ Plus In Vitro DNA Transfection Reagent adds a proprietary booster peptide. SignaGen puts the gain at up to 20-fold over the earlier version in transfection efficiency, across a wide span of mammalian cell types. Gene delivery with GenJet™ Plus has been demonstrated in numerous established cell lines and in primary cells too — HEK293, 293T, 293E, CHO, COS-1, HeLa, NIH/3T3, the insect lines Sf9 and Sf21, plus many other eukaryotic cell types — with cytotoxicity staying low.

A single 1.0 mL vial of the reagent stretches to about 667 transfections in 24-well plates, or 333 in 6-well plates.

Performance Data

GenJet™ Plus DNA In Vitro Transfection Reagent – nuclear GFP in primary rat aortic smooth muscle cells vs L2K
GenJet™ Plus is the top choice for hard-to-transfect cells. Efficiency comparison of GenJet™ Plus In Vitro DNA Transfection Reagent (left panel) with Lipofectamine 2000 (L2K, right panel) on primary cultured rat aortic smooth muscle cells. A nuclear-targeted GFP DNA was transfected with GenJet™ Plus and L2K, respectively per the manufacturer’s protocols. The efficiency was checked 48 hours post-transfection. The above pictures were kindly provided by Dr. Nickolai Dulin of the Section of Pulmonary and Critical Care, University of Chicago
GenJet™ Plus DNA In Vitro Transfection Reagent – lentivirus production and HeLa GFP transfection vs L2K, LTX and FuGene 6
Left Panel: Comparison of GenJet™ Plus In Vitro DNA Transfection Reagent with leading products, including Lipofectamine 2000 (L2K) and Lipofectamine LTX (LTX) on lentivirus production on 293T cells. The data were provided by Dr. Miguel Toscano of Temple University as a courtesy. Right panel: Comparison of GenJet™ Plus In Vitro DNA Transfection Reagent with transfection reagents from other vendors, including Lipofectamine 2000 (L2K) and FuGene 6, on HeLa cells. HeLa cells were transfected with plasmid DNA encoding GFP. Transfections were performed according to the manufacturer’s recommendations and analysed by flow cytometry
GenJet™ Plus DNA In Vitro Transfection Reagent – lentivirus titers from 293T cells vs Lipofectamine LTX by FACSGenJet™ Plus DNA In Vitro Transfection Reagent – lentivirus titers from 293T cells vs Lipofectamine LTX by FACS
Exceptionally high titers of lentivirus were generated from 293T cells. Three DNAs co-transfected with GenJet™ Plus In Vitro DNA Transfection Reagent (left panel) in comparison with Lipofectamine LTX (right panel). 1.0 and 10 microliters of virus supernatant from 293T cells were used to counter-infect 293F cells, which were then passed by FACS. The numbers in the upper right corner indicate the percentage of transduced cells. The titers of lentivirus generated with GenJet™ Plus and Lipofectamine LTX were quantified to be 2.1×106 and 9.3×106 tu/ml, respectively. The above data were provided by Dr. Miguel Toscano of Temple University as a courtesy.
