Scientific Background

Cre recombinase is a site-specific DNA recombinase from bacteriophage P1 that catalyzes recombination between loxP sites, enabling conditional gene activation or deletion in mammalian cells. HEK293T-Cre-eGFP is a stable cell line generated by transducing HEK293T cells with the LV-EF1a-Cre-eGFP-IRES-Puro lentiviral vector (SKU: BHV22000017), followed by puromycin selection. The constitutive EF1a promoter drives robust expression of the Cre-eGFP fusion protein, enabling both Cre recombinase activity and green fluorescence for cell visualization, live-cell imaging, assay development, and experimental optimization. The puromycin resistance cassette allows for efficient antibiotic selection and long-term maintenance of a stable Cre-eGFP-expressing cell population. Each lot undergoes quality control testing to confirm stable transgene expression. Cells are validated through a minimum of 30 serial passages to ensure long-term expression stability, and cryopreserved stocks are tested for viability retention up to 5 years at −80°C. Mycoplasma screening is performed on every lot to confirm negative status.

Usage Notes

Thaw the cryopreserved vial rapidly in a 37°C water bath until a small ice crystal remains. Transfer cells to a pre-warmed culture vessel containing complete growth medium (DMEM supplemented with 10% FBS and appropriate antibiotics). Allow cells to attach for 12–24 hours before changing medium. Maintain selection pressure with puromycin at the recommended concentration (1.0 µg/mL) to preserve transgene expression.

Culture at 37°C in a humidified atmosphere with 5% CO₂. Passage cells at 70–90% confluency using standard trypsinization protocols. Do not exceed passage 20 for optimal transgene expression stability.
